Explainable AI-Driven Identification of Multimodal Biomarkers for Early Prediction of Cognitive Decline
A research group led by Yamaguchi University investigated the prediction of cognitive status over an 18-month period by applying a two-stage machine learning framework with Explainable AI (XAI) to multimodal data from 98 community-dwelling older adults.※
The study integrated diverse data sources—including blood tests, metabolites (including chiral amino acids), gut microbiota, and physical activity recorded via wearable devices—and utilized SHAP (SHapley Additive exPlanations) analysis to visualize the key predictors.
The findings revealed distinct biological and behavioral patterns at different stages of cognitive decline:Progression from healthy to mild impairment: Associated with renal/metabolic functions, amino acid balance, and variability in physical activity.
Progression to more severe stages: Suggested the involvement of changes in energy metabolism, oxidative stress, and choline pathways.As further validation proceeds, these results are expected to pave the way for low-invasive and highly scalable community-based cognitive screening systems.
